Development of an α-synuclein positron emission tomography tracer for imaging synucleinopathies
Synucleinopathies are characterized by the accumulation of α-synuclein (α-Syn) aggregates in the brain. Positron emission tomography (PET) imaging of synucleinopathies requires radiopharmaceuticals that selectively bind α-Syn deposits. We report the identification of a brain permeable and rapid washout PET tracer [18F]-F0502B, which shows high binding affinity for α-Syn, but not for Aβ or Tau fibrils, and preferential binding to α-Syn aggregates in the brain sections. Employing several cycles of counter screenings with in vitro fibrils, intraneuronal aggregates, and neurodegenerative disease brain sections from several mice models and human subjects, [18F]-F0502B images α-Syn deposits in the brains of mouse and non-human primate PD models. Quality difference between Fritillariae Thunbergii Bulbus and Fritillariae Hupehensis Bulbus based on HPLC-ELSD fingerprints and multi-component quantitative analysis
The HPLC-ELSD fingerprints of Fritillariae Thunbergii Bulbus (FTB) and Fritillariae Hupehensis Bulbus (FHB) were established to compare the differences between the two herbs combined with multi-component quantitative analysis. The chromatographic column was Waters ACQUITY HSS T3 (4.6 mm× 250 mm, 5 μm). The gradient elution was performed at a flow rate of 1.1 mL/min with acetonitrile-0.1% triethylamine solution as the mobile phase. The column temperature was 38℃. Evaporative light scattering detection was used. The HPLC-ELSD fingerprints of FTB and FHB were established. The differences between FTB and FHB were compared by chemometrics methods and simultaneous determination of five alkaloid components. The third Intensive Care Bundle with Blood Pressure Reduction in Acute Cerebral Haemorrhage Trial (INTERACT3): an international, stepped wedge cluster randomised controlled trial
Background
Early control of elevated blood pressure is the most promising treatment for acute intracerebral haemorrhage. We aimed to establish whether implementing a goal-directed care bundle incorporating protocols for early intensive blood pressure lowering and management algorithms for hyperglycaemia, pyrexia, and abnormal anticoagulation, implemented in a hospital setting, could improve outcomes for patients with acute spontaneous intracerebral haemorrhage.
Methods
We performed a pragmatic, international, multicentre, blinded endpoint, stepped wedge cluster randomised controlled trial at hospitals in nine low-income and middle-income countries (Brazil, China, India, Mexico, Nigeria, Pakistan, Peru, Sri Lanka, and Viet Nam) and one high-income country (Chile). Higher Chern-Simons based on (2-) crossed modules
We present higher Chern-Simons theories based on (2-) crossed modules. We start from the generalized differential forms in Generalized Differential Calculus and define the corresponding generalized connections which consist of higher connections. Then we establish the generalized Chern-Simons forms to get the higher Chern-Simons actions. Finally, we develop the higher second Chern forms and Chern-Weil theorems.
